Giant uses XS-XL sizing on both the TCR and Defy with their compact sloping top tube - virtual top tube is what matters, not seat tube length. Pick the same letter size on both bikes for your height; the difference is in stack and reach rather than overall size. At Large the Defy stacks roughly 35mm taller with a slightly shorter reach than the TCR, so the bars sit higher without needing a stack of spacers. Riders between two Giant sizes should size up on the Defy for endurance comfort and down on the TCR for a racier drop. Standover is generous on both thanks to the compact geometry.
Choose the TCR Advanced if you race, climb hard or want a fast, aggressive position on smooth roads. Choose the Defy Advanced if you ride long, rough or mixed-surface roads and want a more upright, comfortable platform.
