Two endurance road bikes from two American giants, both with their own takes on vibration damping. The Trek Domane uses IsoSpeed - an active decoupler at the seat tube and head tube. The Cannondale Synapse uses SAVE - micro-flex zones built into the frame and seatpost. Trek's solution is more mechanical and more effective on heavy chip-seal. Cannondale's is simpler, lighter, and cheaper to live with. Both clear 35 mm tires (Synapse) or 38 mm (Domane). Most buyers will be happy on either.

The Synapse sits taller in stack at every comparable size - a Synapse 54 (575/373) is notably more upright than a Domane 54 (571/380) and reaches less. Riders crossing from Domane to Synapse will find a more relaxed fit at the same nominal cm size. At the small end, the Synapse 44 (530/365) is the smallest endurance bike either brand offers - genuinely useful for riders under 5'2" who cannot fit a Domane 49. At the top, the Synapse 61 (640/395) is close to the Domane 61 (645/405) but with a noticeably shorter reach.
If you ride rough roads regularly or want the option of light gravel, the Trek Domane is the more capable bike - IsoSpeed is more effective than SAVE on heavy vibration and the extra 3 mm of tire clearance matters. For long, smoother road days, the Synapse gives you 80 % of the comfort for thousands less, and the SAVE design has no service items to worry about. The Synapse is the smarter all-rounder spend. The Domane is the better tool for genuinely rough surfaces.
Trek Domane - IsoSpeed plus the extra 3 mm of tire clearance wins on rough surfaces, though SAVE is closer than the spec sheet suggests
Cannondale Synapse - lighter frame and quicker handling on smoother roads, with meaningful savings at every price tier
